The Effect of Juglandis Semen of Oxidant - induced Impairment of Membrane Transport Function in Renal Tubules




Abstract
This study was undertaken to determine whether Juglandis semen (JA) exerts beneficial effect aganist oxidant-induced inhibition of p-aminohippurate (PAH) uptake which is actively secreted by renal proximal tubules. PAH uptake increased as function of incubation time to 60 min. when renal cortical slices were exposed to 1 mM tBHP, PAH uptake was significantly inhibited. The inhibition was significantly protected by addition of 5% JA extraction. the protective effect of JA was cose-dependent over the concentration range of 0.1-5%; tBHP (1 mM)-induced inhibition of PAH uptake was completely protected by the concentrations highter than 0.5% JA. PAH uptake was dependent on the medium pH. The inhibitory effect of tBHP and protective effect of JA were not altered by changes in medium pH. tBHP (1 mM) markedly increased lipid peroxidation, which was completely prevented by 5%JA.
These results suggest that JA exerts protective effect against oxiant-induced impairment of membrane transport function and that this effect may be due to by an antioxidant action.
